Standardization of electrolyte leakage data and a novel liquid nitrogen control improve measurements of cold hardiness in woody tissue

Abstract Background A variety of basic and applied research programs in plant biology require the accurate reliable determination tissue cold hardiness. Over past 50 years, electrolyte leakage method has emerged as a popular practical for quantifying amount damage inflicted on by exposure to freezing temperatures. Numerous approaches carrying out this analyzing resultant data have emerged. These include multiple systems standardizing modeling raw protocols boiling or autoclaving samples order maximize positive control. We compare four different routines standardization assess novel control method—immersion liquid nitrogen lieu traditional autoclaving—and apply them woody twigs collected from 12 maple ( Acer ) species early spring. these using each previously published forms analysis vs. controls validate against visual estimates differential thermal analysis. Results Through presentation our own re-analysis findings, we show that both minimum maximum attainable allows estimation hardiness at level across studies diverse systems.
